ESPRESSO microscopy : faster and deeper hysperspectral imaging
Lorenzo Scipioni & Francesco Fersin
Chemoresistant populations are rare subpopulations (0.1 – 4% of the cell population), making it difficult to characterize and follow their behavior. Furthermore, the most biologically relevant in vitro models, tumor spheroids and organoids, are challenging to image due to their 3D organization. To solve these problems, we develop a new hyperspectral encoding system to increase the current imaging speed >400x, while using adaptive optics and structured illumination to achieve higher penetration and resolution, enabling the fast and precise analysis of complex biological samples. At the same time, we develop and implement a suite of machine learning to enhance image quality, automate processing and analysis to perform phenotypic classification based on multidimensional profiles
